Type III secretion systems (T3SS) are widely distributed, essential virulence determinants of Gram-negative bacteria. T3SS serve to translocate, upon contact with eukaryotic host cells, proteins effectors of virulence from the bacterial cytoplasm into the host cytoplasm. Our laboratory studies the function and structure of the T3SS of Shigella flexeri, the agent of bacillary dysentery, using a variety of cell biological, biochemical, genetic and structural approaches. We specifically focus on the energising ATPase, the needle and the translocation pore of the T3SS system, as well as on the function of some putative but still uncharacterised secreted effectors within host cells.
